报告人简介:

Geijsen博士目前担任荷兰皇家科学院首席教授,乌得勒支大学再生医学教授,iTOP技术联合创始人和NTrans公司创立者。他的主要研究成果包括4篇Nature,2篇Cell,6篇Cell Stem Reports,5篇 Cell Stem Cell,5 篇 EMBOs等多篇文章,其中10篇被引数超过100次,3篇被引用200多次。


内容介绍:

The safe delivery of CRISPR technology in patient cells is an important hurdle that needs to be overcome to bring these technologies to the clinic. In recent years, our lab has developed a novel technology, termed iTOP, to deliver recombinant CRISPR/Cas9 ribonucleoprotein (RNPs) complex into patient cells (D’Astolfo et al. 2015 Cell 161, 674-690). We will present how this technology and its potential application in developing immune therapies to treat specific cancers. In addition, we will show recent data demonstrating that iTOP delivery can be applied to deliver CRISPR/Cas9 RNPs in vivo as well, providing interesting new possibilities to treat genetic disease.

In addition to their application in transplantation therapy, stem cells also find application in the selection of treatment options in patients with rare- or difficult to treat diseases such as genetic disease or cancer. We will demonstrate how intestinal organoids, 3-dimensional structures that preserve the cellular anatomy of the intestinal epithelium, can be applied to predict clinical the outcome of drug regimens to treat Cystic Fibrosis or used to screen for new drugs.
